Down Syndrome, or Trisomy 21, results from a chromosomal abnormality where an extra copy of chromosome 21 is present. Statistics indicate that 60% of Down Syndrome cases result in miscarriage during early fetal development, while survivors often face intellectual disabilities, distinct facial features, growth disorders, and multiple anomalies. Medical confirmation has shown that the incidence of Down syndrome is related to the mother's gestational age, with older pregnant women and aging eggs being the main causes of Down syndrome.

Dr. Nathan Zhang, an expert in the field of assisted reproduction and a medical Ph.D. graduated from the University of Pennsylvania said that after in vitro pregnancy and normal prenatal examinations, mothers who choose IVF in American can rest assured because the technology of IVF in American is currently very advanced, which can accurately and effectively screen for 125 negative diseases and chromosomal diseases. However, Dr. Zhang pointed out that despite initial Next-Generation Sequencing (NGS) testing during American IVF, some diseases, like Down Syndrome, may still manifest during fetal development. Down Syndrome is not a result of prenatal development but is present from the early stages of embryo development. While NGS testing involves extracting four cells when the embryo has developed around 128 cells, and these cells might appear normal, it does not guarantee the entire embryo's normalcy, a phenomenon known as chromosomal mosaicism. Dr. Zhang explained that, despite the 99% accuracy of sample testing, a more accurate assessment requires testing 10, 15, or even 20 cells due to the 1% uncertainty.

Moreover, not all diseases can be conclusively identified. Currently, only known genetic diseases can be detected through NGS testing. There are genetic diseases with unknown causes that NGS cannot identify. Additionally, developmental diseases may emerge during the embryo's development, even if it appears healthy during transplantation. Such diseases can only be detected through ultrasound examinations during pregnancy and fetal development.
